Overview
The Zirconium Ultra micro- and nano-UHPLC pump of the Swiss independent vendor Prolab Instruments has been a big success in many application fields such as metabolomics, drug discovery, proteomics, and lately, single-cell proteomics with ultra-low nanoflow requirements. Following our users' requests for a more integrated system for low-flow applications, Prolab has developed an autosampler to complement the Zirconium pumps - the Zirconium CUBE. The Zirconium system now forms a complete HPLC front-end for all low-flow and high speed applications - leveraging the pump's unmatched range of low nl/min to high μl/min flow rates and the autosampler's unique integration features to minimize dead volumes, contaminated surfaces, waiting time, and overall complexity.
A Host of Innovations 
When the sample won't come to the injector, the injector has to go to the sample: Unique on the market, the Zirconium CUBE autosampler is built around a mobile injector unit that includes the injector valve and loop, the sample aspiration needle, as well as the needle wash station. This whole assembly is moved to each sample vial.
The uncommon design approach has a row of stunning advantages: By the rigid connection between the needle and the sample loop, the dead volume and contaminated surface are minimized, and it is possible to use a range of different materials: Flexible fused silica or PEEK capillaries for lowest sample volumes or biocompatibility requirements, stainless steel, but also titanium can be used for the needle as well as for the loop.
Another obvious advantage of the concept is the cycle time saved: The needle does not need to be moved to an injector port nor a wash station. And with the wash station sitting right on top of the needle,sample stains never get to dry before they are immediately washed off.
A Host of Innovations
A key innovation of the CUBE autosampler is its mobile wash unit which cleans the needle between sample injections. It all happens during the time the needle needs to move in and out of the sample vials. No time is wasted moving to a separate wash position, so the stains get no chance to dry on the needle before they are washed off.
High speed and cleanliness do not contradict each other any more!
Minimizing Dead Volume
Dead volume is critical in low-flow HPLC since it equals dead time. Minimizing it directly translates to reduced cycle times and higher throughput. Leveraging the closed-loop flow control algorithm of the Zirconium binary pump, the mixing of two eluents can take place very close to the injector valve, reducing the delay time for the analytical gradient from 20 minutes to 15 seconds in a 300 nl/ min example application.
The delay volume between injector and column and ion source can be minimized when using a special single-rack software configuration, where the injector is fenced to a limited rectangle in the front-right corner of its moving range, allowing the use of the shortest possible transfer capillaries.
Using integrated columns+spray tips further contributes to the reduction of dead volume after the separation, thereby not only saving time but also improving the sharpness of peaks
